Teniposide (VM26) and etoposide (VP16-213) [
E0675] are semisynthetic derivatives of podophyllotoxin [
P1771], an alkaloid. Teniposide and teniposide are an inhibitor of topoisomerase II such as doxorubicin [
D4193], daunorubicin [
D4532]. Topoisomerase II is an enzyme essential for DNA replication, chromosome condensation and chromosome segregation. Topoisomerase II inhibitors are potent inducers of double strand breaks in DNA, and can cause arrest in the cell cycle at the G2 stage. Therefore, topoisomerase II inhibitors are the most widely used anti-cancer agents. Teniposide is used for the treatment of acute lymphocytic leukemia (ALL).
