Tissue-Clearing Reagent iTOMEI-D [for Plants] (1) is a decoloring solution used for the improved TOMEI method (iTOMEI), a transparency method that is an improved version of the TOMEI method developed by Matsunaga et al.1,2) iTOMEI has the advantage of making plants transparent while suppressing the quenching of fluorescent proteins such as GFP and tdTomato. Plants that have been fixed in 1% paraformaldehyde / PBS can be immersed in the decoloring solution 1 to remove chlorophyll from plants. The transparent plants can then be observed by treating them with a mounting solution that matches the refractive index of the objective lens to make the refractive index uniform.
