Application
Leflunomide: A Prodrug of Immunoregulatory Agent, Teriflunomide
Leflunomide is a prodrug of teriflunomide [T3287] which has two activites: inhibition of pyrimidine nucleotide synthesis and interference with tyrosine phosphorylation. More specifically, it inhibits the enzymic activity of dihydroorotate dehydrogenase (DHO-DHase) which catalyzes the fourth step in the biosynthesis of pyrimidine, and tyrosine kinases which transfer a phosphate group from ATP to a protein in a cell. Leflunomide reduces the proliferation of T and B immune cells that are active in multiple sclerosis (MS). For your reference, [C3217] is a synthetic intermediate.
