Guanfacine hydrochloride, a phenylacetyl-guanidine derivative, is a centrally acting α2A-adrenoceptor agonist with a mechanism of antihypertensive action similar to that of clonidine [
D1353]. It reduces blood pressure in human with essential hypertension (EH) at least as effectively as clonidine or methyldopa [
D1817]. Recent research has found that guanfacine can improve prefrontal cortex (PFC) function by strengthening PFC network connections via inhibition of cAMP-potassium channel signaling in postsynaptic spines. Guanfacine is now being used to treat a variety of PFC cognitive disorders, including attention deficit/hyperactivity disorder (ADHD). (The product is for research purpose only.)
