Lurasidone: An Atypical Antipsychotic Agent with the Effects of Antagonizing D2 and 5-HT2A Receptors
Lurasidone (SM-13496) is an azapirone derivative and an atypical antipsychotic agent in development for the treatment of schizophrenia and bipolar depression. In vitro studies has suggested that lurasidone is an antagonist with high affinity for dopamine D2 receptors and serotonin 5-HT2A and 5-HT7 receptors. It also binds with moderate affinity to the human α2C adrenergic receptors to, is a partial agonist at serotonin 5-HT1A receptors, and is an antagonist at the α2A adrenergic receptors. Meanwhile, lurasidone exhibits little or no affinity for histamine H1 and muscarinic M1 receptors. The precise pharmacological mechanism of action of lurasidone is unknown, but lurasidone is believed to show the psychotropic effect through a combination of D2 and 5-HT2A receptor antagonism.
