Trilostane: A Reversible and Competitive Inhibitor of 3β-Hydroxysteroid Dehydrogenase (3β-HSD)
Trilostane is a reversible and competitive inhibitor of 3β-hydroxysteroid dehydrogenase (3β-HSD), which is the key enzyme essential for synthesis of cortisol and all other steroids. 3β-HSD has two isozymes, type1 (3β-HSD1) and type2 (3β-HSD2). 3β-HSD1 is a critical enzyme in the conversion of DHEA [D0044] to estradiol [E0919] in breast tumor cells and may be a target enzyme for inhibition in the treatment of breast cancer, whereas 3β-HSD2 participates in the production of cortisol and aldosterone in the adrenal gland.1,2) Trilostane is an inhibiter of the 3β-HSDs (type 1 and 2). Therefore, trilostane is of interest for the treatment of breast cancer in postmenopausal women2), and has been used for the treatment of Cushing's syndrome in dogs to reduce cortisol and aldosterone levels.3,4) (The product is for research purpose only.)
