We are proud to present the condensation reaction of 4-bromobenzoic acid and morpholine using the capsule reagent. This reagent can omit weighing the condensing agents. 0.25 mmol of HOTU, a condensing agent is encapsulated in each capsule.
Reagents for condensation capsule (HOTU) (one capsule) was added to a solution of 4-bromobenzoic acid (50 mg, 0.25 mmol, 1.0 eq.), morpholine (27 μL, 0.31 mmol, 1.25 eq.) and N,N-diisopropylethylamine (85 μL, 0.50 mmol, 2 eq.) in DMF (1 mL) at room temperature and the mixture was stirred at 30 °C for 2 hours. After the reaction, the mixture was diluted with ethyl acetate (15 mL) and filtered through Celite to remove HPMC. The filtrate was washed with sat. Na2SO4 aq. (10 mL), 2 mol/L HCl aq. (10 mL), sat. NaHCO3 aq. (10 mL) and brine (10 mL). The organic layer was dried over sodium sulfate and concentrated under reduced pressure. The crude product was purified by silica gel chromatography (hexane:ethyl acetate = 2:1 - 1:1) to afford (4-bromophenyl)(morpholino)methanone (54 mg, 80% yield) as a white solid.
