Xanthane Hydride: A Sulfur-Transfer Reagent for Synthesis of Oligonucleotide Phosphorothioates
Xanthane hydride (3-amino-1,2,4-dithiazole-5-thione), Beaucage Reagent [
B3125] and Bis(phenylacetyl) disulfide [
B3623] are used as a sulfur-transfer reagent for synthesis of oligonucleotide phosphorothioates via the phosphoramidite method.
1-4) In particular, xanthane hydride has advantages because it does not yield oxidizing substances, carbon disulfide and cyanamide as the additional reaction products.
2,3) For DNA/RNA synthesizer, xanthane hydride solution is used, such as 0.02 M solution in acetonitrile/pyridine (9:1),
