Application
A Condensation Reagent for Hindered Carboxylic Acids and Low-nucleophilic Amines
Experimental procedure: 2-Methyl-2-phenylpropanoic acid (0.250 g, 1.52 mmol), 4-aminobenzonitrile (0.234 g, 1.98 mmol) and N-methylimidazole (0.42 mL, 5.33 mmol) in acetonitrile are added with chloro-N,N,N',N'-tetramethylformamidinium hexafluorophosphate (0.517 g, 1.83 mmol) in a single portion. After reaction, the reaction mixture is diluted with isopropyl acetate and water. The layers are separated, the aqueous layer is extracted with isopropyl acetate and the combined organics are washed with water, dried wover magnesium sulfate. The mixtute is filtered and concentrated, and then is purified by silica gel chromatography (heptane/isopropyl acetate) to give N-(4-cyanophenyl)-2-methyl-2-phenylpropanamide (374 mg, 93% yield) as a white solid.
