Description

  • Propidium iodide (PI) is a fluorescent dye that selectively stains cells with compromised membranes, making it a reliable marker for dead cells (Excitation: 488 nm / Emission: 617 nm). This product is provided as a ready-to-use solution, eliminating the need for dilution or weighing. Simply add it directly to cultured or suspended cells to achieve selective staining of dead cells. The stained cells can then be used in various applications, including cell death assays and fluorescence microscopy.

Storage

  • Store at 4 °C and protect from light.

Directions for Use

  • Staining conditions are dependent on your specific application. The following protocol describes a method for suspension cells. Please adjust conditions as needed according to your specific cell type and experimental goals.
  • Caution: Because PI is mutagenic, always wear gloves and goggles, and follow your institution's safety guidelines for handling and disposal.
    1. Collect target cells and centrifuge briefly (300 × g, 5 minutes).
    2. Resuspend in PBS and adjust cell concentration to 105–106 cells/mL.
    3. Add PI solution (Product No. P3406) to a final concentration of 1 to 5 μg/mL.
    4. Incubate at room temperature in the dark for 5 to 15 minutes.
    5. Observe cells on a plate or slide. (Ex: 488 nm / Em: 617 nm)
      ‡ Stained cells can be observed immediately. To reduce background fluorescence, wash cells briefly with PBS before observation.
