Application
Reduction of Ethanethiol Esters to Aldehydes (Fukuyama Reduction)

Typical procedure: A 200 mL flask is charged with Cbz-L-phenylalanine ethanethiol ester (20 g), 10 % Pd/C (3.1 g) and acetone (58 mL) under an argon atmosphere. To the suspension is added triethylsilane (13.9 mL) over 1 h at rt. The completion of the reduction can be checked by TLC. The solvent is carefully removed on a rotary evaporator. The residue is suspended in diethylether (60 mL) and the catalyst is filtered off through a Celite pad and rinsed with diethylether (6 x 20 mL). The filtrate is concentrated under reduced pressure to give a crude oil, which solidified upon addition of hexane (30 mL). The solid is collected by filtration and washed with cooled hexane (7 x 10 mL) to give Cbz-L-phenylalaninal (11.6 g, 70.3 %).
