Application
AgOTf/CuI–Co-catalyzed Tandem Alkynylation–Cyclization Reaction of Ｎ-Iminoisoquinolinium Ylide
Typical procedure (entry 1: R1 = Me, R2 = H, R3 = Ph, R4 = p-MeOC6H4): A mixture of 4-methyl-N'-[5-methyl-2-(phenylethynyl)benzylidene]benzenesulfonohydrazide (77.7 mg, 0.2 mmol) and silver triflate (5.1 mg, 0.02 mmol) in anhydrous 1,2-dichloroethane (0.5 mL) is heated at 70 °C for 1 h. The mixture is then cooled to room temperature. Copper(I) iodide (3.8 mg, 0.02 mmol) and a solution of DBU (91 mg, 0.6 mmol) and 1-(bromoethynyl)-4-methoxybenzene (63.3 mg, 0.3 mmol) dissolved in 1,2-dichloroethane (2 mL) are added subsequently. The mixture is stirred at 70 °C overnight. After the reaction, the solvent is evaporated. The residue is diluted with EtOAc (10 mL), washed with H2O (10 mL), and dried over anhydrous MgSO4. Evaporation of the solvent followed by purification on silica gel column chromatography provides 2-(4-methoxyphenyl)-9-methyl-5-phenylpyrazolo[5,1-a]isoquinoline as a white solid (58.3 mg, yield 80 %).
