Strain-Promoted Inverse Electron-demand Diels-Alder Cycloaddition (SPIEDAC) Reaction with Tetarazine

N-(4E)-TCO-L-lysine is an unnatural amino acid (UAA) which can be incorporated in proteins through genetic code expansion using a modified pair of tRNA/tRNA synthetase (tRNA/RS). N-(4E)-TCO-L-lysine has a strained alkene moiety, and reacts with tetrazines through strain-promoted inverse-electron-demand Diels-Alder cycloaddition (SPIEDAC) very rapidly. According to Plass et al. the reaction between N-(4E)-TCO-L-lysine and TAMRA-tetrazine occurred at k = 35000±3000 M-1 s-1.1)
