Pemetrexed: A Multitargeted Antifolate
Pemetrexed is an antimetabolite that inhibits at least three enzymes involved in the folate pathway. These enzymes are thymidylate synthase (TS), dihydrofolate reductase (DHFR), and glycinamide ribonucleotide formyltransferase (GARFT) which are used in purine and pyrimidine synthesis. Therefore, pemetrexed prevents the formation of DNA and RNA which are required for the growth of both normal cells and tumor cells. Pemetrexed has demonstrated clinical activity in non-small cell lung cancer and mesothelioma as well as in a broad array of other solid tumors. For your reference, [A2618] and [M2426] are key synthetic intermediates of pemetrexed. (The product is for research purpose only.)
