Application
Troglitazone: An Agonist at the Peroxisome Proliferator-Activated Receptor of the Gamma Subtype (PPARγ)
Troglitazone (CS-045), a thiazolidinedione such as rosiglitazone [R0106] and pioglitazone [P1091 (HCl salt)], is an agonist at the peroxisome proliferator-activated receptor of the gamma subtype (PPARγ), a transcription-regulating receptor.1) PPAR receptors are found in peripheral tissues important for insulin action. Activation of PPARγ nuclear receptors modulates the transcription of a number of insulin responsive genes involved in the control of glucose and lipid metabolism. Therefore, troglitazone decreases insulin resistance in the periphery tissues and in the liver resulting in increased insulin-dependent glucose disposal and decreased hepatic glucose output. In addition, the PPARγ-independent antitumor effects of troglitazone has been reported.2) Troglitazone was developed as the first in the thiazolidinedione class of hypoglycemic agents, but it was later withdrawn from the pharmaceutical market due to hepatotoxicity.3,4) (The product is for research purpose only.)
