Description
- Green I Nucleic Acid Stain is a highly sensitive fluorescent dye used to detect double-stranded DNA in agarose and polyacrylamide gels. It fluoresces upon binding to DNA. It has an excitation maximum at 490 nm with secondary peaks at 260 and 370 nm and a fluorescence maximum at 520 nm, making it similar enough spectrally to fluorescein that it is compatible with most common filter sets used in imaging equipment. Detection using UV and visible illuminators is also possible. Additionally, it offers higher detection sensitivity than ethidium bromide. This product is supplied as a 10,000-fold stock solution in dimethyl sulfoxide (DMSO). For staining, dilute it 10,000-fold with TAE buffer or a similar solution before use.
Storage
- Store at -20 °C. This product is photosensitive and should be protected from light. Avoid repeated freezing and thawing. Use promptly after opening.
Direction for Use
- Perform electrophoresis using an agarose or non-denaturing polyacrylamide gel.
- Dilute this product (Product No. N1391) 10,000-fold with TAE, TBE, or TE buffer.
Note: Use a buffer with a pH between 7.5 and 8.0 (preferably 8.0) for dilution. Use the diluted solution within 24 hours. The diluted staining solution can be used up to three times. Use polypropylene containers. - Cover the gel with the staining solution and gently agitate it at room temperature for 10 to 30 minutes.
Note: Protect the staining container from light by covering it with aluminum foil or placing it in a dark location. The staining time varies depending on the thickness of the gel and the ratio of agarose to polyacrylamide. No destaining is necessary. - Detect the fluorescence using an imager.

Detection is also possible when using a UV transilluminator with a wavelength of either 250 nm or 300 nm. Using a 250 nm transilluminator enables higher sensitivity detection.
