Research on regenerative medicine
CHIR 99021 is also used with XAV 939 as a key component for mouse epiblast stem cell and human embryonic stem cell self-renewal.1) Under cytokine-free conditions, the combination of CHIR 99021 and the mTOR inhibitor, rapamycin, maintains mouse and human hematopoietic stem cells.2) In the presence of CHIR 99021, OCT4-transduced human blood CD34+ cells are converted into induced mesenchymal stem cells (iMSCs).3)

CHIR 99021 is a specific glycogen synthase kinase 3 (GSK3) inhibitor. GSK3 is a serine/threonine kinase that mediates the addition of phosphate molecules onto serine and threonine amino acid residues, and that plays a key role in both the insulin and Wnt signaling pathways.1,2) CHIR 99021 specifically inhibits the GSK3 pathway.3,4)
