Spiramycin: A 16-Membered Macrolide Substance Having Antibacterial Activity
Macrolide antibiotics are classified by the size of the macrocyclic lactone ring of aglycone as 14-, 15- or 16-membered ring macrolides. Macrolides are characterized by similar chemical structures, mechanisms of action and resistance, but vary in the different pharmacokinetic parameters, and spectrum of activity. Spiramycin is a 16-membered macrolide substance having antibacterial activity isolated from the growth of Streptomyces ambofaciens. The antibacterial spectrum of spiramaycin is quite broad from Gram-positive and Gram-negative bacteria to Toxoplasma gondii, a parasitic protist. Spiramycin inhibits protein synthesis by binding to bacterial 50S ribosomal subunits with an apparent 1:1 stoichiometry.
