WST-8 reagent comprises a pre-prepared, ready-to-use solution which enables the relative quantification of cell number upon addition to cell culture vessels via the detection of intracellular reductive species such as NADPH, NADH, and FADH2. Intracellular redox homeostasis is critical for a wide variety of physiological processes, and as such, cells of the same clone (cell line) are considered to have similar levels of these reductive species.1) When reduced, WST-8 converts from a colorless species to an orange species (Amax = 460 nm), where the degree of color change is proportional to the concentration of reductive species present (reduction potential).2,3) However, as WST-8 has poor membrane permeability, an intermediate molecule is required. This intermediate, the cell permeable electron carrier 1-mPMS, is used to shuttle electrons between intracellular reductive species and extracellular WST-8, allowing for its color change. As this color change is directly proportional to the total amount of reductive species present, and therefore, via induction, the number of cells present, by comparing samples with/without treatment and/or over a set time period, the effect of treatment on cell numbers (cell viability) as well as the degree of cell proliferation can be quantified.
