Cell Culture-Ready Antibiotic Stock Solution

Penicillin-streptomycin solution is a pre-prepared solution containing penicillin and streptomycin, two antibiotics commonly used to prevent bacterial contamination in animal cell culture, sold as a 100x solution for ease-of-use. Penicillin is a β-lactam antibiotic which exerts its antibacterial effect though the inhibition of the peptidoglycan synthase PBP.1) Peptidoglycan is a major component of the bacterial cell wall, and as such is required for both the maintenance and proliferation of bacterial cells.2) While not specific to either gram-positive or gram-negative bacteria, penicillin shows a stronger effect against gram-positive bacteria, whose exposed cell wall leaves them particularly dependent on its integrity. Streptomycin is an aminoglycoside antibiotic which directly binds to the 16S rRNA present in the 30S subunit of bacterial ribosomes, rendering them unable to synthesize peptides.3) As a result, it strongly inhibits the proliferation of both gram-positive and gram-negative bacteria.
