CUBIC-L, X1, and X2 for Observation of Biological Samples Using Expansion Microscopy

Expansion microscopy is an experimental technique reported by Boyden et al. in 2015. In this method, biological samples are fixed, forming a hydrogel, and then physically expanded by taking advantages of changes in solvent properties, which allows for easier observation.1) With CUBIC tissue-clearing reagents, biological samples can be cleared with expansion by simple sequential immersion in CUBIC-L, CUBIC-X1, and CUBIC-X2, without gel fixation. Using this approach, a mouse brain expands to approximately 2.2 times its original length2) and about 10 times its original volume.3) Tissue clearing by expansion microscopy using CUBIC reagents enables observation of biological samples while preserving intracellular structures without destruction.3)
