Valuable Fluorinated Compounds

One of the major trends in recent organic reaction development is the introduction of fluorine atom. Because fluorine is the most electronegative element of all elements, the overall property of a given molecule can be changed to great extent by introducing fluorine atom(s). The atomic radius of fluorine is similar to that of hydrogen at the same time, so the size of the molecule is not affected as much. Due to these properties, the number of compounds containing fluorine is increasing in pharmaceutical drugs and organic electronics materials and so is the demand for effective fluorination reactions.

Generation of the First 3,4-Piperidyne and its Cycloaddition Reactions for the Synthesis of Annulated Piperidines

  Garg et al. have reported the generation of the first 3,4-piperidyne and its cycloaddition reactions for the synthesis of annulated piperidines. According to their results, silyl triflate 1 is treated with CsF in the presence of several trapping agents to afford the corresponding cycloaddition products via 3,4-piperidyne intermediates. For example, in the case of using N-tert-butyl-α-phenylnitrone as a trapping agent, the cycloaddition reaction proceeds regioselectively to afford the isoxazoline product 2 in good yields. Since the compounds bearing piperidine scaffolds are efficient for the synthesis of pharmaceuticals, this reaction is expected to be a new synthetic method for those compounds.

An Asymmetric Organocatalyst Having an Isothiourea Moiety

HBTM-2.1 (1) is an isothiourea derivative having two asymmetric carbons and is used as an asymmetric organocatalyst.1-4) 1 can be used as a catalyst for condensations of two different carbonyl components and consequently affording cyclic compounds by intramolecular cyclizations. For example, an acyl iminium cation intermediate 2 is generated by the reaction of 1 with an anhydride prepared from a carboxylic acid and pivaloyl chloride, followed by the asymmetric 1,4-addition with an unsaturated carbonyl compound giving an enolate intermediate 3. The formed intermediate 3, having an active amide moiety, is cyclized by the intramolecular cyclization from the enolate moiety to afford the desired 6-membered lactone derivative.1) The condensation reactions using 1 have wide availabilities because various cyclic compounds can be synthesized by replacing the unsaturated compounds with other ones. Especially, the synthesis of β-lactam derivatives using imines is considered as one of the efficient conversions.2)
