[Featured Product] Umemoto's Reagent II -A Powerful Electrophilic Trifluoromethylating Reagent

The trifluoromethyl group has unique properties such as high electronegativity, metabolic stability and lipophilicity. The installment of a trifluoromethyl group is one of the significant functionalizations and is utilized for development in various fields, like pharmaceuticals, agricultural chemicals and material science. Umemoto's Reagent II [D5335] is useful electrophilic trifluoromethylating reagent, and applicable for many reactions under basic conditions such as trifluoromethylation of β-ketoesters at the α-position, direct trifluoromethylation of indoles at the 2-position, and hydroxytrifluoromethylation of styrene derivatives using photoredox catalysts.

Nitroxyl-radical-catalyzed Oxidative Coupling of Amides with Silylated Nucleophiles

Moriyama et al. have reported the nitroxyl-radical-catalyzed oxidative coupling of amides with silylated nucleophiles. According to their results, in situ generated N-halogenated amides from amides and a halogenation reagent are activated by the treatment of a nitroxyl-radical TEMPO catalyst to afford the corresponding imine intermediates, which further react with silylated nucleophiles giving the desired coupling products in high yields. This reaction opened up new frontiers in nitroxyl-radical-catalyzed reactions, so it is expected that nitroxyl radicals can be used as organocatalysts in various transformations.

Trifluoromethylthiolating Agent Usable for Direct Dehydroxytrifluoromethylthiolation of Alcohols

Silver(I) trifluoromethanethiolate (1) has already been known as a trifluoromethylthiolating agent1,2) and is also applicable to direct dehydroxytrifluoromethylthiolation.3) 1 successfully gives trifluoromethanethiolate (2) by treatment with tetrabutylammonium iodide (TBAI) or potassium iodide.4) 2 is unstable and is known to decompose into carbonothioic difluoride (3) and fluoride anion. 3 reacts with alcohols to form an intermediate 4. Then, 4 reacts with 2 to afford the corresponding trifluoromethylthiolated product. The trifluoromethylthiolation by using 1 can be applied to alcohols with other functional groups and heterocyclic compounds, so that it is expected to be used in medicinal chemistry.
