Reagents for Introduction of the Teoc-Protecting Group

Typical Procedure:
To a stirred suspension of the amino acid (1 mmol) in water (1 mL) is added a solution of triethylamine (0.21 mL, 1.5 mmol) in dioxane (1 mL) followed by Teoc-OBt (0.31 g, 1.1 mmol) or Teoc-OSu (0.29 g, 1.1 mmol). The mixture is stirred at room temperature overnight, then diluted with water (5 mL), acidified with saturated potassium hydrogen sulfate solution, and extracted with diethyl ether (3 × 15 mL). The combined organic layers are washed with water (4 × 20 mL), dried with magnesium sulfate, and evaporated. The resultant Teoc-amino acid is purified by crystallization (if solid) or obtained as their cyclohexylammonium salts (if they are oils).
