An Efficient Synthesis of Phenanthridines via a Transition-Metal-Free Process


Typical Procedure (R=R'=H):
2'-Bromo-[1,1'-biphenyl]-2-carbonitrile (103 mg, 0.4 mmol) is dissolved in THF (2 mL) in a screw-capped vial (10-mL). The vial is kept stirring for 5 min. Then, Li(Et)3BH (1.0 M in THF, 52 μL, 0.44 mmol, 1.1 eq.) is injected into the reaction mixture. The vial is sealed with a cap, and allowed to stir at 100 °C for 48 h. After completion of the reaction, the reaction mixture is cooled to room temperature and diluted with CH2Cl2 (15 mL), filtered through a Celite pad, and concentrated in vacuo. The residue is purified by flash column chromatography (eluent: hexane/EtOAc = 23/2) to give phenanthridine as a white solid (66 mg, 92%).
