Description
- BSA Maleimide Conjugate is designed to facilitate the conjugation of the BSA to proteins, peptides and ligands that contain free sulfhydryl (-SH) groups. Activated BSA contains 15–35 maleimide groups for each BSA molecule. BSA is conjugated to haptens and typically used as a non-relevant carrier in an ELISA for measuring specific anti-hapten antibody titers.
Protocol
-
- Firstly return the vial to room temperature.
- Reconstitute the contents of the vial with 100 μL of buffer or water to obtain a 10 mg/mL solution of BSA Maleimide.
Note: Do not vortex or heat the BSA Maleimide. - Dissolve 1 mg of a thiol-containing molecule in 100-250 μL of buffer or water. (4-10 mg/mL)
Note: In case of thiol-containing molecules insoluble in buffer or water, DMSO may be used for solubilization (caution: less than 30% DMSO in the final reaction solution). We recommend that the thiol-containing molecule is dissolved in the pH range 6.5-7.5 buffer. 0.1 M sodium phosphate, 0.15 M NaCl, 0.1 M EDTA at pH 7.2 is commonly employed. Maleimides react with sulfhydryls at pH 6.5-7.5 to form stable thioether bonds. EDTA may help to prevent metal catalysed oxidation of thiols. For complete and efficient conjugation, the BSA Maleimide should mix with excess ratio of thiol-containing molecule. Alternatively, if an excess of thiol-containing molecule is not available, it is better to add a sulfhydryl-containing compound, such as cysteine, for quench remaining active maleimide groups. - Immediately mix the thiol-containing molecule and BSA Maleimide solutions and allow them to react for 2 hours at room temperature.
- If desired, desalt, dialyze or purify the conjugate.
Storage
- Store at −20 °C to −80 °C (Preferably at −80 °C). Reconstitute this reagent immediately before use. Do not prepare stock solutions for storage. Discard any unused reconstituted reagent.
