Keratanase II cleaves the sulfated N-acetylglucosaminidic β1-3 linkage to galactose included in keratan sulfate. The enzyme essentially requires the sulfation at 6-position of the constitutive N-acetylglucosamine. On the other hand, it acts regardless of the sulfation at 6-position of the galactose linked to the sulfated N-acetylglucosamine. The Keratanase II gene is derived from Bacillus circulans and its recombinant protein is expressed in Escherichia coli.
Unit Definition
One unit (U) of Keratanase II is defined as the amount required to produce 1 umol of reducing oligosaccharides from shark cartilage keratan sulfate polysaccharide per minute at 37°C under the condition of pH6.0.
Assay Conditions
Reaction conditions: 20 mM sodium acetate, pH6.0
Reaction mixture includes the enzyme and shark cartilage keratan sulfate in 50 ul. The mixture is incubated at 37°C for 15 min. Quantification of reducing end of released oligosaccharides is measured by the modified Somogyi-Nelson method (see reference).
Storage
Store at -20°C. Avoid repetitive freezing and thawing. After thawing the product, it’s recommended to store at 4°C and use it within a few weeks.
