Mitoxantrone Dihydrochloride: A Synthetic Anthracycline Antitumor Agent with Improved Tolerability
Mitoxantrone [
M3133] and its dihydrochloride are synthetic anthracycline antitumor agents that are structurally similar to doxorubicin hydrochloride [
D4193] and daunorubicin hydrochloride [
D4532]. Mitoxantrones intercalate into deoxyribonucleic acid (DNA) through hydrogen bonding, causes crosslinks and strand breaks. .Mitoxantrones also inhibit DNA repair by inhibiting topoisomerase II which results in fragmentation of DNA. Mitoxantrones display an improved tolerability profile compared with doxorubicin and other anthracyclines, but cross-resistance with the anthracyclines has been demonstrated.
