To a solution of N-acetyl-L-phenylalanine (1.0 g, 4.78 mmol) in DMF di-tert-butyl acetal (6.9 mL, 28.7 mmol, 6.0 eq.) was stirred at 60 °C for overnight. The solvent was removed under reduced pressure and the residue was purified by column chromatography (ethyl acetate:hexane = 1:1 on silica gel), giving tert-butyl acetyl-L-phenylalaninate as a colorless oil (1.20 g, 94% yield).

Application
Esterification Reagent for GC Derivatization
Experimental procedure: Fatty acid (~ 5 mg) is placed into a vial and then N,N-dimethylformamide di-tert-butyl acetal (100 μL) is added. The reaction is completed upon dissolution. The reaction mixture can be injected directly into GC.
