Description
- Histones are essential proteins found in the nucleus of eukaryotic cells. They bind to DNA, packaging it into chromosomes and regulating gene expression.
- This product can be used to isolate histones from cultured mammalian cells and tissues, supporting epigenetic analysis.
Directions for Use
- Dilute 10x Lysis Buffer with purified water to prepare 1x Lysis Buffer.
- Collect cells via centrifugation and remove supernatant. Use a cell scraper to detach adherent cells.
- Resuspend cells in cold 1x Lysis Buffer, adding 1 mL for every 1 x 107 cells.
- Incubate on ice for 10 minutes.
- Centrifuge at 3,000 x g for 5 minutes at 4 °C and remove the supernatant.
- Resuspend the pellet in cold 1x Histone Extraction Buffer. Use 1/10 the amount of buffer used in step 3.
- Incubate on ice for 30 minutes.
- Centrifuge at 12,000 x g for 5 minutes at 4 °C.
- Transfer the supernatant (containing histone) to a new tube.
- Add the same amount of 1x Neutralization Buffer as the 1x Histone Extraction Buffer used in step 6.
