Step-by-Step Protocol
- Incubate tissue sections as normal, first with primary antibody, followed by HRP-conjugated secondary antibody. (Preliminary testing of primary antibody concentrations between 1:100 and 1:2000 is recommended.)
- Before first use, dissolve a tyramide reagent in 50 µL DMSO. (Aliquot as needed. Store below -20°C. Minimize freeze-thaw cycles.)
- After secondary antibody staining, wash sections with TBST (5 minutes, 3 times).
- Immediately before use, mix the following to prepare the staining solution for each 1 cm2 area to be stained.
- Tyramide reagent solution (from Step 2) 1 µL
- 0.15% H2O2 1 µL
- 1x TBS 98 µL
- Remove slides from TBST and tilt onto a tissue to remove excess buffer. Lay slides flat, immediately apply 100 µL of the staining solution from Step 4 to each section, and allow to react for 1-10 minutes.
- After the reaction, remove excess solution with a tissue and immediately wash with an excess of TBST (5 minutes, 3 times).
- After washing, apply mounting solution and coverslip for observation.
