Coupling Activators for Oligonucleotide Synthesis

Phosphoramidite chemistry using coupling activators has become dominant in the synthesis of oligonucleotides, due to its high coupling efficiency and quick turnaround time. TCI offers a lineup of various modified activators, including imidazolium salts with different acidities, which can be used to strike a good balance between the activation of phosphoramidite and reducing undesired detritylation (de-DMT group). For example, imidazolium salts like 1-methyl-1H-benzimidazol-3-ium trifluoromethanesulfonate (= N-MeBIT) [M3384], tetrazoles such as 5-(benzylthio)-1H-tetrazole (= BTT) [B3020], and pyridinium salts such as pyridinium trifluoroacetate (= PyยทTFA) [P2920] are available.
