1-Adamantanamine Hydrochloride: A Dopamine Receptor Agonist and an Anti-Influenza A Virus Agent
		
		1-Adamantanamine hydrochloride and its free base [
A2318] are dopamine receptor agonists and anti-influenza A virus agents, similarly rimantadine hydrochloride [
R0070]. 1-Amantadine was originally developed as an antiviral agent. One day, a single patient noticed relief in her Parkinson’s disease symptoms after taking amantadine for a flu infection, and this observation led to a new drug indication.
1)These mechanisms of action are not fully understood. However, there is evidence to suggest that 1-adamantanamine act as a dopamine receptor agonist by enhancing the synthesis and release of dopamine from nerve cells and delaying the reuptake into synaptic vesicles.
2,3)In addition, 1-adamantanamine inhibits the replication of influenza A viruses by preventing the release of infectious viral RNA into the host cell nucleus by interfering with the function of the transmembrane of the viral M
2 protein.
