• Size : 5 mL
• Sterility : Filter-sterilized

Description

• Stains RNA in bacteria.
• T4436 contains T-Fluor 480 dissolved in PBS at a concentration of 800 µg/mL.
• Wavelengths of RNA stained with T-Fluor 480: λ_ex,max = 450 nm, λ_em,max = 485 nm.
• Very high fluorescence intensity. The signal can be observed without washing.
• Tested for RNA staining.

Directions for Use: RNA staining of E. coli with T-Fluor 480

• 1. Cultivate E. coli BW25113 on LB medium at 37 °C overnight.
  2. Add 1/100 volume of culture medium to LB medium with or without 100 µg/mL rifampicin (RNA synthesis inhibitor) and incubate at 37 °C for 4 hours.
  3. Dilute T4436 125-fold with PBS.
  4. After removing the supernatant by centrifugation, add the 125-fold diluted T-Fluor 480 solution to the pellet and stain at room temperature for 5 minutes.
  5. Observe under a fluorescence microscope using a filter for GFP.

Directions for Use: Observation of RNA in transparent biofilm

• 1. Incubate Methicillin-resistant Staphylococcus aureus (MRSA) in a glass-bottomed dish for 24 hours to form a biofilm.
  2. Remove the culture medium and suspended bacteria. Then fix and wash the biofilm.
  3. Dilute T4436 100-fold with PBS.
  4. Add the 100-fold diluted T-Fluor 480 solution to the dish and stain the biofilm at room temperature for 30 minutes.
  5. Add PBS or biofilm clearing reagent iCBiofilm-H1 (Product No. T4031) to the dish and observed the biofilm.

Storage

• This product is delivered frozen but can be stored at 4 °C. Please use it within six months after refrigeration.
