Description
- Biofilms are highly organized structures of microbial communities composed of a matrix of polysaccharides and other substances secreted by microbes and/or aggregates of microbial cells. Microbial cells become resistant to antimicrobial drugs and host defense systems when they form biofilms.
In order to observe the structures of biofilms with a thickness of more than several tens of micrometers, Clearing Reagent iCBiofilm-H2 has been developed. iCBioflim-H2 is supplied as a stock solution. Before adding to biofilm, iCBiofilm-H2 has to be diluted with deionized water to appropriate concentration (undiluted iCBiofilm-H2 may work well). By adding iCBiofilm-H2 solution to a biofilm, the biofilm becomes transparent within several seconds and easy-to-observe by light microscopy such as confocal laser scanning microscopy.

Storage
- Stored at room temperature and used within 6 months.
Direction for Use

- Prepare a biofilm in a glass bottomed culture dish (35mm diam.). Culture conditions (including medium, temperature, time, etc.) all play roles in biofilm formation; the conditions specific to your microbe of interest must be determined separately.
- After removing planktonic cells and culture medium, add 1% glutaraldehyde solution in PBS and fix at room temperature for 30 minutes. To preserve immunogenicity for immunofluorescence applications, we recommend substituting 4% paraformaldehyde in PBS.
- Remove these glutaraldehyde solution and wash three times with PBS or distilled water.
- Stain using appropriate fluorescence reagents.
- Dilute iCBiofilm-H2 (Product No. T4032) with deionized water to an appropriate concentration.
Recommendation ratio; iCBiofilm-H2 : water = 2 : 1 or 1 : 1
If you need the best performance of iCBioflim-H2 for a target biofilm, please consider to examine different concentrations of iCBiofilm-H2. - Remove staining reagents and add diluted or undiluted iCBiofilm-H2 solution (about 500 µL – 1 mL).
- Observe the biofilm by optical microscopy.
